Depending on the research question, we use various diagnostic tools to analyse microbial activity.
qPCR-analysis
qPCR is a quantitative method for measuring DNA and RNA, allowing you to determine the abundance of a pre‑selected target organism or process. Unlike culture‑based methods, qPCR also detects non‑cultivable micro‑organisms (approximately 99% of what occurs in nature). This provides a realistic and reliable picture of microbial activity in pipes, installations and water systems. A key advantage is that samples can be fixed in the field, ensuring that the microbiological composition remains unchanged during transport.
vPCR – analysis of viable micro‑organisms
vPCR is similar to qPCR, but focuses specifically on viable micro‑organisms: cells that are still active or capable of becoming active again, such as spores. The method is always carried out in conjunction with qPCR and reveals which proportion of the micro‑organisms in a sample is still truly ‘active’. Because vPCR — just like qPCR — analyses genetic material, you are not limited to what can be cultured. However, samples for vPCR must be supplied fresh, as they cannot be fixed.
Next-generation sequencing (NGS)
NGS is a tool we use to identify all the micro‑organisms present in a sample, including species that are not yet known. It does not provide exact quantities, but it does show which groups are more or less abundant. Using 16S amplicon sequencing, we map the microbial population and gain insight into the role these micro‑organisms play in a process. The results can be used, for example, to develop qPCR analyses for monitoring. With total RNA sequencing, we can even analyse all organisms and their activity without having to select a target in advance.
Next-generation sequencing with the Illumina MiSeq i100
For our NGS analyses, we use the Illumina MiSeq i100, one of the latest compact NGS platforms. This system delivers high‑quality data, reliable DNA profiles and fast turnaround times. As a result, we can provide clients with in‑depth microbial analyses and well‑founded reports in a timely manner.
